Sexual arousal results in excitatory signals to the nervous system resulting in a release of nitric oxide in the corpus cavernosum (i.e., spongy erectile tissue of the penis). Nitric oxide diffuses into the smooth muscle cells lining the arteries of the corpus cavernosum, thereby relaxing vascular and trabecular smooth muscle cells. The relaxation of the smooth muscles lining the penile arteries allows these arteries of the corpus cavernosum to become engorged with blood, compressing the veins that drain blood from the penis and, as a result, trapping blood within the corpus cavernosum to produce an erection.

The physiology of the male erectile tissue provides numerous points of interaction in which nervous or chemical manipulation may be used to overcome sexual dysfunction or enhance sexual responsiveness.

Nitric Oxide And L-arginine Connection

The enzyme arginase regulates the L-arginine bioavailability to nitric oxide synthase by hydrolyzing L-arginine, thereby depleting the intracellular pool of L-arginine available to form nitric oxide. Therefore, the administration of L-arginine, arginase inhibitors and stimulants of nitric oxide synthase production have been used to enhance male sexual response and to treat sexual dysfunction in males through the enhanced production of nitric oxide. Of these treatments, the administration of L-arginine has been the most extensively studied and successful.

We also know the administration of L-arginine is very effective in increasing the blood in erectile tissue leading to increased sexual arousal. These effects are mediated through the nitric oxide pathway by the increased intracellular supply of L-arginine available for conversion of nitric oxide.
